Yes, the Peloponnese is one of the best regions in Greece for a road trip. The distances between the highlights are manageable and the roads are mostly in good condition.
For a road trip, definitely. Many of the most beautiful places, beaches and mountain villages are very hard, or impossible, to reach without your own car.
Good bases are Nafplio for the east, Monemvasia or the Mani for the southeast, Kalamata for the southwest, Pylos or Koroni for Messenia, and Olympia or Arcadia for the west.
From Athens you can easily reach the Corinth Canal, ancient Corinth, Mycenae, Epidaurus and Nafplio, all doable as day trips within a manageable drive.
April, May, June, as well as September and October are ideal. In high summer it can get very hot, especially inland, while the coast stays mild even outside the main season.
